Study on Chinese Herb Medicine of Anti-Avian Influenza Virus and Variation of M2 Gene Induced by Amantadine Hydrochloride

Avian influenza(AI) is an infection disease by type A influenza virus, and it is classified Ginfection disease by O.I.E. It is great economic loss in poultry industry. The great characteristics of the highly pathogenic avian influenza is high mortality, the rate of death was 100%.In HongKong,H5Nl subtype of avian influenza virus(AIV) was isolated from 18 human and H9N2 subtype of avian influenza virus was isolated from 2 human directly, which was the first time that AIV overacrossed interspecies barrier and infected human.Amantadine hydrochloride is widely used to treat flu. But drug resistance of influenza virus will happen. The M2 gene of 5 strains avian influenza virus isolated in Chinese were amplified by RT-PCR and analysed the sequence and predicted secondary structure of the transmembrane domain. In addition, three amino acid residues 27, 31,34 in the transmembrane region mutated. Secondary structure of the transmembrane domain changed and predicted the function will be changed.Chinese tradition medicine is heritage of Chinese. Not only its source is plentiful, but also its toxic is low. Because its action is several targets, it can solve the drug resistance. To choice the effective Chinese tradition medicine of anti-avian influenza virus , Radix Scutellariae, Radix Astragali, Rhizoma Coptidis, Fructus Forsythiae, Fructus Forsythiae, Flos Lonicerae, Foliom hatidis, Hreba Hoattuyniae and several complex Chinese tradition medicine were selected in vitro and MDCK.The study of in vitro and in vivo showed that the compound medicine had a strong action of anti-avian influenza virus in vivo, it could prolong the mean death time of chicken and took effect on the chicken challenged the subtype H5N1 of highly avian influenza virus.The study of acute toxic experiments on mouse was as following: the compound medicine was safe and the maximum tolerant dose was 400 times. After intraperitoneal injection administration, the LD50 was 425.6mg/kg.b.w.According to the method of residue medicine, the pharmacokinetics of the compound medicine was studied after intraperitoneal injection administration. Ka was 4.9078 or 6.4089h-1, kel was 0.1501 or 0.19634. The parameter showed that the slower its elimination, the lower its concentration. It had a long time in the mouse and it had long effect in the mouse. It could strengthen the immunity function of SPF chicken through promoting maturity of immunity organ. It could promote the ability of anti-stress, increase the percentage ofCD3+,CD/,CDg+,TCR] +T cells in peripheral blood of chickens and increase the function of immunity.
