Experimental study on the treatment of Acute Spinal Cord Injury in rats using Chinese Medicine

Spinal Cord Injury (SCI) is a very serious complication of the illness and fracture of spinal column. These years, from experimental study of postinjury spinal cord in pathopbysiology, we can know that acute traumatic spinal cord injury is seldom complete-transection. The recovery of injured nerves?function is impeded because of the secondary damage, and the prognosis of this disease becomes worse. The mechanism of the progressive injury is not yet understood. It has made a very progress on the study of the mechanism of secondary injury of SCI, accompany with the developing of experimental techniques and checking methods. It's mainly on the subjects of lipid hyperoxidation, active vascular factors, and apoptotic cell. These studies give us therapeutic hints to this disease. There is no such name as SCI and no description associate with this disease in ancient books of Traditional Chinese Medicine (TCM). It should belong to the categories of "ei Syndrome" in TCM according to its clinical manifestations. Pathogenesis of this disease is the injury of the Du Channel, basing on the location of the injury and physiology of the Du Channel. It interferes with the dysfunction of Channels and Collaterals, Zang-Fu, Qi and Blood, et al. It generally regards blood stasis as the foundation in pathology, so the therapeutic principle is to promote blood circulation and remove blood stasis, in order to clear the Du Channel in the early stage of SCI. My tutor based on the pathogenesis of SCI as well as his rich clinical experiences and investigation of modem pharmacology formulated ji-Zhui-Zhi-Liao-Fang. The clinical study about this decoction showed it could improve the function of CNS. Adult Sprague-Dawley rats injured by modified Allen's method acted as the models in these experiments. The therapeutic protocol was Methylprednisolone (MP). These parameters, the effect of anti-lipid hyperoxidation, influence to active vascular factors, morphological changes, as well as influence to functional recovery, were investigated after cord injury in rats treating with TCM. Based on these findings, the efficacy of TCM and the mechanism of its effect, is discussed. 1.Changes in the activity of superoxide radicals using TCM after experimental spinal cord injury. TCM and MP can promote the functional recovery of spinal cord in 7, 14 days postoperation significantly, according to behavioral analysis of functional neurologic impairment (combined behavioral score, CBS). There was no difference between TCM treated group and MP treated group in 7 days postoperation, but the efficacy of TCM was much significant in 14 days postoperation. Furthermore, TCM had less side effects and could be used for a long time. It provided the theoretical basis for the conclusion of having beneficial effects and less side effects of this decoction in clinical study. There was no difference between TCM treated group and MP treated group about MDA changes of spinal cord in 6 and 24 hours postoperation, but TCM had stronger effect on curbing the increasing of MDA containing in 14 days postoperation. The SOD containing of TCM treated group, did not change so much in 6h and 24h postoperation, increased significantly and was closed to normal level in 14 days postoperation.
