Study on the treatment mechanism of experimental endometriosis model in rat using Chinese medicine

Endometriosis is a common and debilitating woman problem, the pathogenesis of endometriosis has not been understood yet up to now. It most frequently affects women in the reproductive years, and is uncommon prior to the menarche. The lesions will atrophy after the menopause. Medical therapy and oophorectmy make a low estrogen condition, which reduce the number and size of lesions, but some lesions recurrence following estrogen return to normal after medical therapy is over. These clinical observations highlight the importance of hormonal factors in the pathogenesi S of endometr ios is. It is thought as a estrogenepended disease. There is no such name as endometriosis in ancient books of Traditional Chinese Medicine (TOM). It should belong to the categories of TongJing(dysmenorrhea)?hengJia (tumor) ~Yu Zi(infertility) ?and ?Yue Jing Bu Tiao(abnormal menstruation)?in TOM according to its clinical manifestations, Its pathogenesis of TOM is blood stasis, so the therapeutic pr inc iple is promot ing blood circu lat ion and remov ing blood stas is, combined with resolving masses or clearing Fu Qi or reinforcing kidney. Luo Shi Nei Yi Fang is formulated by the late famous gynecologist of TOM, Luo yuanai, who had rich clinical experiments in using Chinese medichine for treatment of endometriosis . The decoction can promote blood circulation and remove blood stasis and resolve masses. The clinic study had showed it improved immunology function of patients. Based on these findings, endornetriosis model in rat was used to further explore the treatment mechanism in present study. We observed the alterations of hormone and hormone receptor as well as blood reheology and ulrastructure of ectopic endometrium before and after treatment of Luo Shi Nei Yi Fang decoction. 1.Blood rheology effects of Chinese medicine in the treatment of endometriosis model The results of blood rheology in model control group did not change compared with normal control group, so there had no significant difference between decoct ion treated group and model control group. But we found danazol effected blood rheology significantly, resulting in hyperviscosity Syndrome, which might relate to side effects of danazol . Chinese drugs promoting blood circulation can improve hyperviscos ity Syndrome, so we suggest that endometr iosi s could be treated with western medicine combined with Chinese medicine to increase curative effect and decrease side effects. 2. Endocrine effects of Chinese medicine in the treatment of endometriosis model Serum estradiol (E2) level decreased markedly in danazol treated group, so did prolactin(PRL) level , while follicle梥timulating hormone (FSH) increased . It is concluded that therapeutic actions of danazol is 慳nti梕stradiol 憈hen ectopic endometrium atrophy without E2support. Luo Shi Nei Yi Fang decoction did not decrease but increase serum E2 level ,higher than model control group. Serum F2 level had significant difference between decoction treated group and danazol treated group. The result has a impuls ion that Chinese medicine might play therapeutic actions from different ways in the treatment of endometriosis, and have not so much side effects due to low Eaas danazol has. 3.
